Secreted parasite protein antagonizes TGF-beta signaling in fibroblasts by host receptor interaction
In a collaborative effort, spearheaded by Andy Hinck and coworkers at the University in Pittsburgh, we identified that the secreted helminth TGM6 protein that acts as a potent antagonist of TGF-beta signaling in fibroblasts. TGM6 has adapted its domain structure and sequence to mimic TGF-b binding to the host TGF-beta type II receptor (TbRII). See the remarkable similarity at the interfaces of X-ray crystallography structures of the TGM6-D3:TβRII (A) and TGF-beta/TbRII (B) complexes.
